Macromolecules have hydrophilic and low permeability properties that hinder drug deliverythrough conventional routes, making parenteral administration the most effective but alsomost invasive and least preferred by patients. Permeation enhancers have been shown to increase membrane permeability to facilitate drug permeation across the mucosal epithelium, offering an alternative route for drug administration through the oral transmucosa. This study aimed to compare the concentration dependent effects of dodecyl β-D -maltoside (DDM) and sodium taurocholate (NaTc) in enhancing fluorescein isothiocyanate dextran 4 (FD4) permeability across the porcine oesophageal mucosa. Flow through diffusion cells and fluorescence spectrophotometry were used to quantify FD4 permeation over 24 hours. DDM increased FD4 permeation at 0.5mM, 5mM, and 10mM consistently between biological specimens. NaTc enhanced permeation at 10mM, whereas at 25mM and 50mM outcomes varied between biological specimens. This study showed that DDM demonstrated consistent enhancement of FD4 permeation, while NaTc requires repeating experiments. Incorporating in vivo like conditions in future studies would validate enhancers permeability effect.

Makromolekyler har hydrofila och låg permeabilitetsegenskaper, vilket påverkar läkemedelsadministration via konventionella vägar och gör parenteral tillförsel mest effektiv men samtidigt invasiv och mindre föredragen av patienter. Permeationsförstärkare har visats öka membranpermeabiliteten och därmed underlätta läkemedelstransport igenom mukosala epitelet, vilket erbjuder en alternativ väg för läkemedelsadministrering genom det orala transmukosan. Denna studie syftade till att jämföra de koncentrationberoende effekterna av dodecyl β-D-maltosid (DDM) och natriumtaurokolat (NaTc) på permeabiliteten av fluorescein isotiocyanat dextran 4 (FD4) genom gris eosafagusmukosa. Flödes genom diffusionsceller och fluorescensspektrofotometri användes för att kvantifiera FD4-permeation under 24 timmar. DDM ökade FD4 permeationen vid 0.5mM, 5mM, och 10mM, med konsekvent resultat mellan biologiska specimen. NaTc ökade permeation vid 50mM, medan resultaten vid 25mM och 10mM varierade mellan biologiska specimen. Studien visade att DDM uppvisade en konsekvent permeationsförstarkelse, medan NaTc kräver ytterligare upprepade experiment. Att inkludera in vivo liknande förhållanden i framtida studier skulle stärka bedömningen av permeationsförstärkarnas effekt.
