Gene Expression in Caenorhabditis elegans Fed Bacteria Pre-treated with Metal Contaminated Soil Leachates and Lab Metal Mixture2016Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [en]

The accumulation of heavy metals in the environment mainly from anthropogenic sources can be harmful to organisms. Humans, as well as many organisms are primarily exposed to heavy metals through their food source, and this can have seriously damaging effects. This study used Caenorhabditis elegans to determine the effects of feeding on bacteria exposed to heavy metal soil leachates by analysing their gene expression. The contaminated soils were collected near a glass factory in Sweden and their effect on C. elegans was compared to a lab prepared metal mixture of a similar concentration. Of the genes 16 evaluated, 8 showed a significant response when exposed to heavy metals. Sample site 1 induced up regulation in cdr-1 and cyp-35A2, and down regulation in hsp-70, hsp-16.2, and hsp-16.48. Sample site 2 induced up regulation in ftn-1 and cyp-35A2, and down regulation in mtl-1, hsp-70, hsp-16.2, and hsp-16.48. Sample site 3 induced up regulation in mtl-1, cdr-1, gst-4, and cyp-35A2. Finally, the metal cocktail induced up regulation in only mtl-1 and gst-4. The exposure of C. elegans to heavy metals through their food source did not show any signs of lethality but some of the key genes were certainly affected. This implies that the high levels of metals present in the soil leachates caused gene dysregulation in C. elegans to cope with the resultant stress. The difference between the stress response of the soil leachates and metal cocktail can be attributed to the complexity of the environmental samples which is comprised of several other components apart from metals.

The effects of different browning agents SuperHume, HuminFeed, and Reverse Osmosis Concentrate on the fitness and survival of Daphnia magna2019Independent thesis Advanced level (degree of Master (Two Years)), 30 credits / 45 HE creditsStudent thesis
    Abstract [en]

The browning of freshwater bodies due to increasing concentrations of dissolved organic carbon (DOC) is becoming more prevalent in light of the changing climate and is therefore a relevant topic of study within the scientific community. An obstacle to overcome however, is what to use as a source of carbon for scientific experiments. Reverse osmosis concentrate (ROC) is a natural source filtered from wetlands, though this can take a long time to collect. Other studies have turned to commercial products which are leonardite-based to simulate browning: HuminFeed, which comes as a powder or SuperHume, which is liquid. Here I present the results that test the effects of these browning agents on zooplankton. After a previous study indicated a severe impact on zooplankton in a lake mesocosm experiment, a controlled lab experiment was conducted. The model test organism Daphnia magna and copepods from a local lake were used to observe the effects of HuminFeed, SuperHume and ROC in a standard immobilisation and reproduction test using similar concentrations as the mesocosm experiment. The browning agents did not affect the immobilisation of D. magna or the copepods after 24 or 48 hours, however the reproduction test on D. magna resulted in a significant decrease in the total number of offspring produced in the HuminFeed treatment compared to the control. This study raises the question if the commercial product HuminFeed is an acceptable substance to use to mimic browning in lab or mesocosm experiments

Skolsköterskors erfarenheter av hälsofrämjande arbete med överviktiga barn2014Independent thesis Advanced level (degree of Master (One Year)), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

Metod: Individuella semistrukturerade

intervjuer med åtta skolsköterskor genomfördes. Materialet analyserades med hjälp av

kvalitativ innehållsanalys. Resultat: Analyserna resulterade i fyra kategorier och tolv

underkategorier.

Behavioral and neuroendocrine correlates of selection for stress responsiveness in rainbow trout - a review2005In: INTEGRATIVE AND COMPARATIVE BIOLOGY, Vol. 45, no 3, p. 463-474Article in journal (Refereed)
    Abstract [en]

In rainbow trout the magnitude of the cortisol response to stress shows both consistency over time and a moderate to high degree of heritability, and high responding (HR) and low responding (LR) lines of rainbow trout have been generated by individual selection for consistently high or low post-stress cortisol values. Using 2nd and 3rd generation fish, we tested the hypothesis that differential stress responsiveness is associated with behavioral alterations in the HR-LR trout model. LR fish showed a tendency to become socially dominant, a rapid recovery of food intake after transfer to a novel environment, and a reduced locomotor response in a territorial intrusion test. Furthermore, stress induced elevation of brain stem and optic tectum concentrations of the monoamine neurotransmitters serotonin, dopamine, and norepinephrine and their metabolites suggests that both synthesis and metabolism of these transmitters were elevated after stress to a larger degree in HR than in LR trout. A divergent pattern was seen in the hypothalamus, where LR fish displayed elevated levels of 5-hydroxyindoleacetic acid (a serotonin metabolite) and 3-methoxy-4-hydroxyphenylglycol (a norepinephrine metabolite). Thus, selection for a single trait, cortisol responsiveness, in rainbow trout is associated with concurrent changes in both behavior and central signaling systems. The apparent parallel to genetically determined stress coping styles in mammals, and the existence of similar trait associations in unselected populations of rainbow trout, suggests an evolutionarily conserved correlation between multiple traits. Continuing studies on the HR and LR trout lines are aimed at providing the physiological and genetic basis for new marker-assisted selection strategies in the rapidly developing finfish aquaculture industry, as well as increased knowledge of the function and evolution of central neuroendocrine signaling systems.

    Abstract [en]

The aim of this review article is to present an overview of the, in the Nordic countries, relatively new research area ’children exposed to domestic violence’, and to discuss the implications of this research. The overview shows that the international research is primarily quantitative, focusing on children’s symptoms. Only a small part of the published research studies are qualitative. These studies use the term ‘experience’, rather than ‘exposed’, stressing the child’s subject status. The Nordic research is qualitative, and in line with early North-American studies, uses the term ‘to witness’. Although the field has greatly expanded, the field still have many unanswered questions concerning for example recilience, gender and ethnicity, and lack un understanding of the relational and contextual aspects from the child’s perspective.

    Abstract [en]

The aim of this study is, by analysing children's and young people's discourses, to investigate their strategies in response to domestic violence episodes, in relation to their age. The empirical data come from individual interviews with children and young people (ages 8–20 years) who had experienced domestic violence and lived at refuges for abused women. The thematic analysis shows that the children describe a wide range of strategies before, during and after a violent episode, that all children act regardless of age and that strategies vary according not only to age but also to situation and context. The theoretical framework used is the sociology of childhood, and the analysis engages with theoretical concepts of age, agency and positioning.

Girls on the Verge of Exploding?: Voices on Sexual Abuse, Agency and Sexuality at a Youth Detention Home2004Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

The present thesis investigates the dilemmas and difficulties the staff at a youth detention home encounter and struggle with when working with young women who are understood to be victims of sexual abuse. At the center of attention is talk about the problems of talking about sexual abuse and other difficult experiences. The overall aim in conducting the study was to open up an arena that has been neglected and little investigated - youth detention homes for girls and young women, by talking to staff and the young women at the detention homes, and talking about sexual abuse. Also in focus was the young women's own thinking about the body and sexuality.

The findings are presented in five articles. The first article examines what discursive devices are employed when using the focus group method when talking to the young women in forced care. The study shows in what ways the focus groups is a fruitful method for studying marginalized young women and their views and thoughts about being young women today. The second article addressed the issue of how the staff form narratives of sexual abuse. Stories of sexual abuse were "power stories" as well as "work identity stories" and were considered to have the power to heal as well as the power to harm. The third article examines the process leading up to the definition of sexual abuse. The study is concerned with the process in which the staff members define whether or not a young woman has been a victim of sexual abuse. A determining factor was whether or not the act involved a person who was defined as a victim. A core issue was an evaluation of the credibility of the alleged abused girl and the degree of consent. The fourth article addresses the issue of how the staff and the young women at the detention home talk about sexuality. The article compares the different views of the staff and the young women and concludes that the staff talk about the young women as asexual children and as victims of sexual abuse, and the young women talk about themselves as having sexual agency. Finally, the fifth article shows how the young women talk abut childbearing and motherhood.

The study shows that the issue of talking about sexual abuse and other difficult experiences is complex. The different views of how to talk about sexual abuse, whether to talk about sexual abuse, when to talk and to whom, as well as the question about whether there is a need to talk, makes the issue of talking about sexual abuse multi-layered and contradictory.

Innocent Girls or Active Young Women?: Negotiating Sexual Agency at a Detention Home2003In: Feminism and Psychology, ISSN 0959-3535, E-ISSN 1461-7161, Vol. 13, no 3, p. 345-367Article in journal (Refereed)
    Abstract [en]

This article analyzes discourse about sexuality at a detentionhome for young women aged 13-21 years. Staffmembers, both male and female, talk about the young women asif they were still children, i.e. as asexualbeings, thereby denying them sexual agency. Theimage of young women as innocent children is, however, contestedby the young women themselves, who explicitlyclaim sexual agency. The article concludes thatby not using the situatedness of a young woman as their therapeuticalpoint of departure, the staff members cannotprovide her with the guidance and support that she needs. The article further suggests that feminist theoryshould focus on young women's own perceived senseof agency and that sexual agency can be understood and established in the context of Moi's (1999) model ofwomen as `de Beauvoirian' situated beings.

Samtal från ett särskilt ungdomshem2006In: Socialvetenskaplig tidskrift, ISSN 1104-1420, Vol. 13, no 2, p. 164-178Article in journal (Refereed)
    Abstract [sv]

Våld i hemmet - barns strategier2012Book (Other academic)
    Abstract [sv]

    Abstract [en]

In recent years, the interest in including children and adolescents in research on violence and abuse has increased. According to the UN Convention on the Rights of the Child, children have the right to be heard in issues that concern them and their lives. However, this puts great demands on researchers and the projects they design. This article discusses challenges in shedding light on children’s experiences and including them as informants. The starting point for discussion is three research projects aimed, in different ways, at measuring or exploring the dynamics of children exposed to violence. Using these studies as a backdrop, three main questions are asked. What con-cepts are used to describe children’s experiences and why is the choice of concepts important? Do our surveys measure what we think we measure, and do our interviews capture what we want to capture if our aim is to explore and understand children’s lifeworld? What are some of the central ethical dilemmas researchers face when conducting research on children and violence? Finally, the importance and implications of these methodological and empirical challenges are discussed.

    Abstract [en]

Using data from a study on police officers' encounters with domestic violence victims and a study on children experiencing domestic violence, this article examines how officers decide whether and how to communicate with children in emergency situations, and how children experience these encounters. Officers' views on such communication diverge; usually, communication is motivated by the need to determine next actions. Children recall little communication and describe officers as faceless, nameless and genderless. The authors argue for recognizing the preventive role of officers on emergency calls. Official policies and guidelines should formally acknowledge and clarify the importance of communication with children.

    Abstract [en]

This article discusses whether the focus group method can be employed with troubled groups and for the discussion of high‐involvement topics. It analyses focus groups' discourse of high‐involvement topics, such as ‘the body’, ‘relationships’, and ‘sexuality’, conducted with female adolescents aged 15–20 years at a detention home. Contrary to the traditional belief that the focus group method is designed for low‐involvement topics and mainstream groups, our analyses of the discursive devices employed suggest that the method can indeed be used for high‐involvement topics. We also argue that other methods would not have given us ‘deeper’ insights and that the focus group method can be seen as a less intrusive method to be used in this setting.

‘If you look, you have to leave’: young children regulating research interviews about experiences of domestic violence2014In: Journal of Early Childhood Research, ISSN 1476-718X, E-ISSN 1741-2927, no Aug 4, p. 1-13Article in journal (Refereed)
    Abstract [en]

The aim of this article is to investigate the competence of young children staying with their mothers in refuges for abused women as participants in qualitative interviews. Discourse of the verbal and non-verbal actions of seven young children (4–7 years old) was analysed using a theory originally developed to describe infant–mother interaction as a model. The analysis shows that the young children were able not only to communicate important aspects of what it means for a child to live in a family with domestic violence but also to regulate, limit and take the lead in the interviews, similar to the ways infants regulate their internal states during interactions with their caretakers. The findings emphasize the importance of including children this young in research and challenge taken-for-granted notions of adult power and helpless children.

The aim of this article is, by analysing childrens discourses, to investigate their actions or absence of actions during a domestic violence episode. The empirical data are recorded group therapy sessions and individual interviews with children who have grown up experiencing their fathers violence against their mothers. The analysis shows that the childrens stories contain two aspects of actions: one related to the actions during the ongoing episode, and one the child perceives as possible/desirable for the future. The findings are discussed in the light of Lazarus and Folkmans theory of coping.

Work identity at stake: The power of sexual abuse stories in the world of compulsory youth care2003In: Narrative Inquiry, ISSN 1387-6740, E-ISSN 1569-9935, Vol. 13, no 1, p. 217-242Article in journal (Refereed)
    Abstract [en]

This article deals with issues of narrative power in the highly contested area of sexual abuse stories. The storytellers are all members of a group of resident assistants working at a Swedish detention home for young women aged 13–21. Being a victim of sexual abuse was seen as a fundamental component in the complex picture of self destructive behavior impairing the young women. Expressing their suffering and talking about these experiences were seen as important aspects of the positive psychological development that was the aim of mandating the young women to compulsory care. The analysis focuses on the performative, destabilized struggle of the staff to maintain their work identity at the detention home, as they talked about their encounters with young women victimized by sexual abuse. The object of examination is how the work identity of the resident assistants at the institution was enacted and lived through in a narrative form as accountable patterns of meaning, as they took an active part in the process of narrating. The stories were “power stories” as well as “work identity stories”, considered to have the power to heal as well as the power to harm, and the staff regarded themselves as responsible for the storytelling, at the same time as they were not in control of either the act of storytelling or its outcome.

Flickor i skolsvårigheter: En fenomenologisk studie om flickor med sen ADHD-diagnos2019Independent thesis Advanced level (professional degree), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

Konsekvenserna kan bli stora ända in i vuxenlivet.

    Abstract [sv]

    Abstract [sv]

Akoella ja susilla on mualima pilattu: Naisen arvostus Suomen kansan sananparsikirjassa2015Independent thesis Advanced level (degree of Master (One Year)), 10 credits / 15 HE creditsStudent thesis
    Abstract [fi]

Assessment of Coverage and Effect of Web Content Accessibility Guidelines 2.12018Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

The Web Content Accessibility Guidelines (WCAG) are an important resource for making the web more accessible for people with disabil- ities. They are used as a source of information for web content cre- ators, but also in standards as a basis for legislation when measuring whether a website is accessible or not. The guidelines have been criti- cized for poor coverage when studying real-life accessibility, especially for people with cognitive impairments. A newly released version of the guidelines, WCAG 2.1 aims to improve accessibility but has not been studied yet.

This study evaluates the coverage of WCAG guidelines by con- ducting a user study and comparing the results with WCAG evalu- ation to find out if the problems found by users would be fixed by conforming to the guidelines. 3 websites were evaluated with a total of 6 users from 3 different disability groups. Same sites were evaluated with WCAG 2.0 and 2.1.

The results show none of the site passed the WCAG evaluation. The users found 40 issues, of which 32,5% would be fixed if conform- ing to WCAG criteria. Most of the issues not covered by WCAG were related to understandability and readability of the content and the user interface. The blind group found the biggest number of issues, but al- most half of the issues would be fixed by WCAG, compared to only 13% for people with cognitive impairments. Despite the fact that all critical issues were covered by WCAG, the amount of serious issues not covered reveals that WCAG as a sole measure for accessibility is not enough to guarantee equal access to the web.

Förskollärares syn på matematik: En studie om hur förskollärare resonerar kring det matematiska arbetet i förskolan2017Independent thesis Basic level (professional degree), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

Characterisation of Members of the HD-Zip I and DREB/ERF Transcription Factor Families and their Functions in Plant Stress Responses2010Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

Plants have to adapt to various environmental conditions. In comparison with other eukaryotes, plant genomes encode relatively many transcription factors, and have families of transcription factors only found in plants, two such are the large AP2/ERF (APETALA2/Ethylene Responsive Factor) and HD-Zip (Homeodomain leucine zipper) families. Members of these families play roles in plant development, responses to stress and other environmental stimuli. This thesis describes the functions of members of both these families. Two AP2/ERF proteins of the DREB/ERF (Drought Responsive Element Binding/Ethylene Responsive Factor) subfamily, AtERF38 and AtERF39 show a negative effect on growth in transgenic over-expression lines. They are regulated by stress and confer stress tolerance, which suggests a function as negative growth regulators during stress. The 14 Oryza sativa (rice) HD-Zip I genes were described and a phylogenetic analysis of the HD-Zip I genes in Arabidopsis thaliana, rice and Craterostigma plantagineum defined four clades that were conserved between mono and dicots, but also showed that four clades had most likely been lost in either lineage. The phylogeny was supported by conserved intron/exon patterning and the duplication history of the rice genome. The expression patterns in different organs indicate a non-conserved regulation of genes within and between clades. Drought stress regulation was conserved within two clades, but not between the clades. Further analysis on the function of four HD-Zip I genes in Arabidopsis, ATHB5, -6, -7 and -12 showed an involvement in the transcriptional regulation of ABA receptors and signalling components. These are the first described direct target genes for ATHB7 and ATHB12. It was also shown that there exists a direct cross-regulation between these four HD-Zips. The effect of ATHB5, -6, -7 and -12 would be as negative regulators of ABA signalling, desensitising the plant to constant levels of ABA, and enabling sensitivity to changes in stress level.

Evaluation of the improvement of rolling analysis by means of modern technology2006Conference paper (Other academic)
    Abstract [en]

Modern wire rod rolling is characterised by high rolling speed and requirements on well-defined mechanical properties as well as close tolerances. Traditional methods for analyses of this kind of processes, such as the slip line field theory and the upper bound analysis, have proven not to be sufficient. Different methods for rolling analysis have been evaluated. Several FE approaches were compared to classical methods such as the upper bound and the slip line theory and the results was validated by practical experiments. The finite element method has proven to be a powerful tool in order understand the metal working process, it also gives the possibility to study phenomena not yet clarified.

The influence of bearing geometry on the residual stress state in cold drawn wire, analysed by the FEM2006In: Journal of Materials Processing Technology, ISSN 0924-0136, E-ISSN 1873-4774, Vol. 171, no 3, p. 446-450Article in journal (Refereed)
    Abstract [en]

Finite element simulations have been performed in order to study the influence of bearing geometry on the residual stress-state in cold drawn wires. Experiments in full industrial scale have been performed in order to verify the FEM results. The residual stresses were measured with X-ray diffraction in both the axial and tangential direction. The material used was a high carbon steel for roller bearings, 100Cr6. It was found that the geometry of the bearing has a large influence of the residual stress-state. When the bearing is somewhat tapered, a gap in the beginning of the bearing is formed. This "double contact" reduces the axial and tangential residual stress to minimum or even to a compressive state. A cylindrical bearing gave no contact in the bearing at all due to elastic deformation. This resulted in a large axial and tangential residual stress. It was shown that even a small divergence from the desired bearing geometry due to the grinding of the die could affect the residual stress immensely.

The interdependence of plastic behaviour and final properties of steel wire, analysed by the finite element method2004Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

The finite element method (FEM) has been used in order to study the interdependence of plastic behaviour and final properties in steel wire. Three processes metal forming have been studied: billet forging, wire rod rolling, and cold drawing.

The forging study focuses on the possibilities to eliminate voids, pores and pipes by altering the bite ratio in open die forging. Forging is used in wire manufacturing when dealing with materials that are difficult or impossible to roll, e.g. tool steels like high-speed steels, and powder metallurgically manufactured materials like tungsten-molybdenum filament wire. The elimination of inner defects by forging were successfully simulated by the FEM.

The rolling study focuses on the problems due of the extreme finish rolling velocities in modern wire rod mills and modern technologies such as thermo-mechanical rolling and sizing. A design for a high-speed laboratory mill for studies of material behaviour at extreme strain rates was made. With a combination of two-high stands and three-roll units is it possible to obtain a smoother temperature distribution in the bar cross section.

Two studies of the cold drawing process were made. The first describes the distribution of the plastic deformation due to the die geometry. It was found that the influence of the die angle on the amount of inhomogeneous deformation decreases with increasing area reduction. The frictional behaviour was also studied.

The second study describes how the bearing geometry affects the residual stress-state in the wire. It was found that a small taper could change the residual stress state in the surface immensely. This was shown with FE simulations and verified by X-ray diffraction measurements. The results in this paper gives a tool for control of the residual stress-state in the wire. This gives a possibility to increase e.g. the fatigue strength in the final product.
