    Abstract [sv]

Membrane protein topogenesis2015Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

The membranes of cells are highly complex and heterogeneous structures that fulfill multiple vital tasks. They form thin barriers that seal out the environment, thus defining the cell’s boundaries. They mediate the selective exchange of information and substances between the inside and outside of cells, thus making cellular life and survival possible and allowing fast adaptation to changing conditions. Not least importantly, they harbor key components of many essential processes such as the photosynthesis and respiration. Membranes are composed of a largely apolar lipid matrix densely punctuated with a large number of different proteins. These so-called membrane proteins usually span the lipid matrix and protrude out into the space on either side of the membrane.

Over millions of years of evolution, cells have developed an incredible machinery to facilitate the insertion of membrane proteins into the membrane. Our understanding of these machines and the insertion processes they mediate has reached a point where we have a very good picture of membrane protein biogenesis in various types of cells. However, more data still needs to be collected to completely comprehend the complex molecular mechanisms and the physical chemistry that underlies the different insertion processes.

The work presented in this thesis contributes to that understanding. More precisely, we have studied how weakly hydrophobic transmembrane elements of membrane proteins, which cannot spontaneously enter the largely apolar membrane matrices, are efficiently incorporated. Indeed, such elements are quite common in membrane proteins and our work has lead to the formulation of a novel mechanism for how they can be integrated into biological membranes.

We have also added to the understanding of the physical chemistry that underlies the membrane insertion process by systematically introducing non-proteinogenic amino acids into a membrane-spanning segment of a membrane protein and studying its membrane insertion capability.

Quantitative Analysis of SecYEG-Mediated Insertion of Transmembrane alpha-Helices into the Bacterial Inner Membrane2013In: Journal of Molecular Biology, ISSN 0022-2836, E-ISSN 1089-8638, Vol. 425, no 15, p. 2813-2822Article in journal (Refereed)
    Abstract [en]

Most integral membrane proteins, both in prokaryotic and eukaryotic cells, are co-translationally inserted into the membrane via Sec-type translocons: the SecYEG complex in prokaryotes and the Sec61 complex in eukaryotes. The contributions of individual amino acids to the overall free energy of membrane insertion of single transmembrane alpha-helices have been measured for Sec61-mediated insertion into the endoplasmic reticulum (ER) membrane (Nature 450:1026-1030) but have not been systematically determined for SecYEG-mediated insertion into the bacterial inner membrane. We now report such measurements, carried out in Escherichia coli. Overall, there is a good correlation between the results found for the mammalian ER and the E. coli inner membrane, but the hydrophobicity threshold for SecYEG-mediated insertion is distinctly lower than that for Sec61-mediated insertion.

Orientational Preferences of Neighboring Helices Can Drive ER Insertion of a Marginally Hydrophobic Transmembrane Helix2012In: Molecular Cell, ISSN 1097-2765, E-ISSN 1097-4164, Vol. 45, no 4, p. 529-540Article in journal (Refereed)
    Abstract [en]

alpha-helical integral membrane proteins critically depend on the correct insertion of their transmembrane alpha helices into the lipid bilayer for proper folding, yet a surprisingly large fraction of the transmembrane alpha helices in multispanning integral membrane proteins are not sufficiently hydrophobic to insert into the target membrane by themselves. How can such marginally hydrophobic segments nevertheless form transmembrane helices in the folded structure? Here, we show that a transmembrane helix with a strong orientational preference (N-cyt-C-lum or N-lum-C-cyt) can both increase and decrease the hydrophobicity threshold for membrane insertion of a neighboring, marginally hydrophobic helix. This effect helps explain the missing hydrophobicity in polytopic membrane proteins.

Apolar surface area determines the efficiency of translocon-mediated membrane-protein integration into the endoplasmic reticulum2011In: Proceedings of the National Academy of Sciences of the United States of America, ISSN 0027-8424, E-ISSN 1091-6490, Vol. 108, no 31, p. E359-E364Article in journal (Refereed)
    Abstract [en]

Integral membrane proteins are integrated cotranslationally into the membrane of the endoplasmic reticulum in a process mediated by the Sec61 translocon. Transmembrane α-helices in a translocating polypeptide chain gain access to the surrounding membrane through a lateral gate in the wall of the translocon channel [van den Berg B, et al. (2004) Nature427:36–44; Zimmer J, et al. (2008) Nature455:936–943; Egea PF, Stroud RM (2010)Proc Natl Acad Sci USA 107:17182–17187]. To clarify the nature of the membrane-integration process, we have measured the insertion efficiency into the endoplasmic reticulum membrane of model hydrophobic segments containing nonproteinogenic aliphatic and aromatic amino acids. We find that an amino acid’s contribution to the apparent free energy of membrane-insertion is directly proportional to the nonpolar accessible surface area of its side chain, as expected for thermodynamic partitioning between aqueous and nonpolar phases. But unlike bulk-phase partitioning, characterized by a nonpolar solvation parameter of 23 cal∕ðmol · Å2Þ, the solvation parameter for transfer from translocon to bilayer is 6 –10 cal∕ðmol · Å2Þ, pointing to important differences between translocon-guided partitioning and simple water-to-membrane partitioning. Our results provide compelling evidence for a termodynamic partitioning model and insights into the physical properties of the translocon.

Energetics of side-chain snorkeling in transmembrane helices probed by nonproteinogenic amino acids2016In: Proceedings of the National Academy of Sciences of the United States of America, ISSN 0027-8424, E-ISSN 1091-6490, Vol. 113, no 38, p. 10559-10564Article in journal (Refereed)
    Abstract [en]

Cotranslational translocon-mediated insertion of membrane proteins into the endoplasmic reticulum is a key process in membrane protein biogenesis. Although the mechanism is understood in outline, quantitative data on the energetics of the process is scarce. Here, we have measured the effect on membrane integration efficiency of nonproteinogenic analogs of the positively charged amino acids arginine and lysine incorporated into model transmembrane segments. We provide estimates of the influence on the apparent free energy of membrane integration (Delta G(app)) of snorkeling of charged amino acids toward the lipid-water interface, and of charge neutralization. We further determine the effect of fluorine atoms and backbone hydrogen bonds (H-bonds) on Delta G(app). These results help establish a quantitative basis for our understanding of membrane protein assembly in eukaryotic cells.

Positional editing of transmembrane domains during ion channel assembly2013In: Journal of Cell Science, ISSN 0021-9533, E-ISSN 1477-9137, Vol. 126, no 2, p. 464-472Article in journal (Refereed)
    Abstract [en]

The integration of transmembrane (TM)-spanning regions of many channels and ion transporters is potentially compromised by the presence of polar and charged residues required for biological function. Although the two TMs of the ATP-gated ion channel subunit P2X2 each contain charged/polar amino acids, we found that each TM is efficiently membrane inserted when it is analysed in isolation, and uncovered no evidence for cooperativity between these two TMs during P2X2 integration. However, using minimal N-glycosylation distance mapping, we find that the positioning of TM2 in newly synthesized P2X2 monomers is distinct from that seen in subunits of the high-resolution structures of assembled homologous trimers. We conclude that P2X2 monomers are initially synthesised at the endoplasmic reticulum in a distinct conformation, where the extent of the TM-spanning regions is primarily defined by the thermodynamic cost of their membrane integration at the Sec61 translocon. In this model, TM2 of P2X2 subsequently undergoes a process of positional editing within the membrane that correlates with trimerisation of the monomer, a process requiring specific polar/charged residues in both TM1 and TM2. We postulate that the assembly process offsets any energetic cost of relocating TM2, and find evidence that positional editing of TM2 in the acid-sensing ion channel (ASIC1a) is even more pronounced than that observed for P2X2. Taken together, these data further underline the potential complexities involved in accurately predicting TM domains. We propose that the orchestrated repositioning of TM segments during subunit oligomerisation plays an important role in generating the functional architecture of active ion channels, and suggest that the regulation of this underappreciated biosynthetic step may provide an elegant mechanism for maintaining ER homeostasis.

Lateral proton transfer between the membrane and a membrane protein.2009In: Biochemistry, ISSN 0006-2960, E-ISSN 1520-4995, Vol. 48, no 10, p. 2173-9Article in journal (Refereed)
    Abstract [en]

Proton transport across biological membranes is a key step of the energy conservation machinery in living organisms, and it has been proposed that the membrane itself plays an important role in this process. In the present study we have investigated the effect of incorporation of a proton transporter, cytochrome c oxidase, into a membrane on the protonation kinetics of a fluorescent pH-sensitive probe attached at the surface of the protein. The results show that proton transfer to the probe was slightly accelerated upon attachment at the protein surface (approximately 7 x 1010 s(-1) M(-1), compared to the expected value of (1-2) x 10(10) s(-1) M(-1)), which is presumably due to the presence of acidic/His groups in the vicinity. Upon incorporation of the protein into small unilamellar phospholipid vesicles the rate increased by more than a factor of 400 to approximately 3 x 10(13) s(-1) M(-1), which indicates that the protein-attached probe is in rapid protonic contact with the membrane surface. The results indicate that the membrane acts to accelerate proton uptake by the membrane-bound proton transporter.

Children with high and intermediate imperforate anus: Remembering and talking about medical treatment carried out early in life2008In: Pediatric surgery international (Print), ISSN 0179-0358, E-ISSN 1437-9813, Vol. 24, no 9, p. 1009-1015Article in journal (Refereed)
    Abstract [en]

Treatment of children with high and intermediate imperforate anus entails several different surgical procedures during the first 3–12 months of life, which are accompanied by a strict follow-up treatment regimen. It has not been studied whether the children remember this treatment carried out early in life. Research has shown that small children may demonstrate so-called non-verbal memories of salient events occurring in early childhood. The purpose was to examine whether children with imperforate anus showed distressing memories of previous medical treatment and whether parent–child dialog about medical treatment is related to the child’s psychosocial functioning later in life. Parents of 25 children (9 boys, 16 girls) with high and intermediate imperforate anus participated in the study. The mean age among the children was 10.5 years (range 8.0–13.6). A comparison group of 30 children (5 boys and 25 girls) with juvenile chronic arthritis also participated in the study. The mean age was 10.6 years (range 7.8–13.6). All parents answered the Child Behavior Checklist (CBCL/4-18) and a study-specific questionnaire. Children in both groups were reported to show distressing memories of early treatment. Children who had been talked to showed good psychosocial function and were in a better mood and less angry than those who had not been talked to. Parent–child discussions about the child’s experiences of medical treatment did not seem to be harmful or in any other way detrimental to the child, instead such discussions seemed to facilitate the child’s psychosocial functioning.

Beyond Democracy in Cambodia: Political Reconstruction in a Post-Conflict Society2009Collection (editor) (Refereed)
    Abstract [en]

For more than three decades Cambodia lived with civil war and genocide. After the peace agreement, major reconstruction efforts and UN-supervised elections in the 1990s, it was hoped that the ravages of the past could be repaired. Instead, one political crisis after another has ensued. And to the extent that some political stability has emerged, seemingly this has been won at the expense of democracy. Moreover, reconstruction efforts appear to be at odds with processes of liberal democratization.

This volume (written by a broad mix of Khmer and non-Khmer researchers) is the first study to assess the post-conflict democratization process in Cambodia in a systematic and in-depth empirical way. In going beyond a one-dimensional view of democracy, the full complexity of this process is illuminated. Not only does the volume focus on the successes and failures of Cambodia’s political elite but also it looks beyond Cambodia, assessing the extent to which the globally applied post-conflict strategy of intervention followed by early elections, hoping for rapid democratization, is sustainable and progressive.

Solid Waste Management Systems and Informal Waste Picking & Recycling Chains in Dhaka, Bangladesh: A Ward-Level Study2011Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

Waste management is a critical issue in the fast growing mega-cities of developing countries today. It is therefore pertinent that studies on all activities contributing to waste management, whether formal or informal, be conducted for decision makers that are actively seeking solutions to problems. The following ward-level study of a neighborhood in Dhaka gleans a three-pronged perspective of the waste-related systems and activities currently in operation. Firstly, the relationship between the formal waste management system and the informal waste picking/recycling chain will be explored, using the systems approach ‘modernized mixtures’ as a framework. Secondly the informal waste picking/recycling chain will be deconstructed, assessing the economic and environmental implications of this chain’s work, against the concept urban symbiosis. Finally, the concept urban symbiosis and it’s ‘proximity principle’ will be used to understand the geographical flow of plastic waste through this informal chain. This study is qualitative, using both empirical data, collected through observation, following (physically trailing behind waste workers on their rounds) and semi-structured interviews conducted in the field, as well as a secondary source literature review. The separate but linked examinations that make up the aims of this study will lend nuanced insights into the waste-related systems at play from a sub-local setting in Dhaka City, contributing both a socio-material systems perspective, an economic and environmental perspective, as well as a geographical perspective, emphasizing the importance of contextual and holistic approaches to municipal waste management by local authorities.

Design and implementation of test a tool for the GSM traffic channel.2002Independent thesis Basic level (professional degree)Student thesis
    Abstract [en]

Todays’ systems for telecommunication are getting more and more complex. Automatic testing is required to guarantee quality of the systems produced. An actual example is the introduction of GPRS traffic in the GSM network nodes. This thesis investigates the need and demands for such an automatic testing of the traffic channels in the GSM system. A solution intended to be a part of the Ericsson TSS is proposed. One problem to be solved is that today’s tools for testing do not support testing of speech channels with the speech transcoder unit installed. As part of the investigation, a speech codec is implemented for execution on current hardware used in the test platform. The selected speech codec is the enhanced full rate codec, generating a bitstream of 12.2 kbit/s, and gives a good trade-off between compression and speech quality. The report covers the design of the test tool and the implementation of speech codec. Particularly performance problems in the implementation of the encoder will be addressed.

Styrelsens Ersättning: Vilka faktorer ligger till grund för denna?2012Independent thesis Advanced level (degree of Master (One Year)), 20 credits / 30 HE creditsStudent thesis
    Abstract [sv]

    Abstract [en]

On January 1, 2014 a legislative amendment to the former National Heritage Act (1988: 950) was put in action, one of the new amendments that was conceived in what is now called Culture Environmental Act (1988: 950). The change made it possible for the County Councils to enforce that the results associated with archaeological research would be conveyed to the public.

The present work is a case study aiming to see what kind of impact this amendment has had on the archaeological specifications and offer invitations for archaeological investigations.

The main questions for the work are:

    • What kind of mediation is being required by the County Councils?
    • Why was the law changed?
    • Is there a difference in this respect between different counties?
    • Has there been any noticable change in practice in this respect after the amendment introduction?

This study aims to answer these questions and throughout it also give you as a reader information about the history behind the changes as well as what impact the changes has had on archaeological investigations and the mediated results to the public. The aim is also to show the different ways that archaelogy is communicated and to whom.

Hur skapar jag ett spel där spelarens huvudsakliga sysselsättning är att måla?: Reflektioner kring arbetet med spelet ”Art Hero”2008Independent thesis Basic level (degree of Bachelor), 20 credits / 30 HE creditsStudent thesis
    Abstract [sv]

    Abstract [sv]

    Abstract [en]

    Abstract [en]

    Purpose

The purpose of this study was to examine the experiences of children with high and intermediate imperforate anus (IA), and specifically their experiences of hospital care.

    Methods

Twenty-five children born with high and intermediate IA participated; 9 boys and 16 girls. The mean age was 10.5 years (range 8.0–13.6). Two control groups were involved in the study. A self-report questionnaire was used to gather the data concerning children’s experiences of hospital care. Items were scored on a 5-point Likert scale.

    Results

The children’s responses on hospital care items scored high. The children with IA reported being less satisfied with the information given, and understood less why they needed to visit the hospital than did the children in the two control groups.

    Conclusion

The children’s experiences of care seemed to be positive even though the children born with IA are subjected to invasive treatment. More research is called for in the unexplored area of information to the children, and particularly to the children born with a defect. Children’s views are important and should always be considered, as their answers most certainly reflect a genuine experience, contributing to the further development of their specific care.

    Abstract [en]

    Background/Purpose

Imperforate anus is an unusual malformation, which, even after surgical intervention, usually entails constipation and fecal incontinence. This study aimed to evaluate ongoing psychosocial effects of this birth defect in school-aged children.

    Methods

Twenty-five children born with high and intermediate imperforate anus participated in the study, along with their parents and classroom teachers. One group of healthy children and 1 group of children with juvenile chronic arthritis, along with their parents, served as controls. Children and parents individually answered a questionnaire devised for this study. Parents filled out the Child Behavior Checklist and the children's teacher filled out the Teacher's Report Form.

    Results

According to test results, children with imperforate anus were happy and optimistic. They liked school better and reported better relationships with schoolmates than the other children. The index group reported statistically significantly more frequent constipation. According to parental responses, the imperforate-anus children suffered from fecal incontinence and odor, as well as constipation (P < .001). Index-group parents reported on the Child Behavior Checklist that their children had more emotional and behavioral problems. On the Teacher's Report Form, teachers reported few problems for the same children.

    Conclusions

Patients with imperforate anus did not experience psychosocial impairment despite significant functional problems.

Parental Experiences: Care of Children With High and Intermediate Imperforate Anus2006In: Clinical Nursing Research, ISSN 1054-7738, E-ISSN 1552-3799, Vol. 15, no 4, p. 290-305Article in journal (Refereed)
    Abstract [en]

In this study parental experiences of care of children with high and intermediate imperforate anus were evaluated. A group of 45 parents of children with high and intermediate imperforate anus and two control groups participated. Data collection with individual questionnaires concerning the child's hospital care, information to the parent and the child, and involvement in the care of the child were performed. Parents of children with imperforate anus reported being less satisfied with the care of their child, and they were less content with information about their child's treatment compared with the control groups. The parents had been extremely involved in the follow-up treatment. Constipation and fecal incontinence are common and involve suffering for the children and their parents. Parents have to be motivated and supportive and have a great deal of patience to be able to put up with caring for these children, and it seems as if health care professionals have underestimated their problems.

    Abstract [sv]

    Abstract [sv]

The size and shape of three water-soluble, non-ionic polysaccharides produced by lactic acid bacteria: A comparative study2016In: Carbohydrate Polymers, ISSN 0144-8617, E-ISSN 1879-1344, Vol. 142, p. 91-97Article in journal (Refereed)
    Abstract [en]

Three water-soluble, non-ionic extracellular polysaccharides (EPS) obtained from lactic acid bacteria (S. thermophilus THS, L. helveticus K16 and S. thermophilus ST1) were subjected to a comparative study by means of multidetector size-exclusion chromatography, providing distributions and averages of molar masses, radii of gyration and intrinsic viscosities. All polysaccharides displayed random coil character. Further analysis of the data reveals differences in chain stiffness and extension that could be well correlated to structural features. The calculated persistence lengths ranged from 5 to 10 nm and fall within the range typical for many single-stranded bacterial or plant polysaccharides. The ST1 polysaccharide had the highest molar mass but the lowest persistence length, which is attributed to the presence of the flexible (1→6)-linkage in the main chain.

    Abstract [en]

Background: During the 2000s Sweden has built so many arenas s that it’s unprecedented in the Swedish history. Billions has been plowed into the arenas while criticism has grown. While this is being written new arenas are planned and built even though almost all new arenas has been proved to be unprofitable. Due to limited research regarding Swedish arenas there’s a need to study and describe the ongoing arena-boom.

Purpose: The main purpose of this study is to increase knowledge about Swedish arena investments regard to the following aspects: Reasons why they have been built, its investment structure and its ownership structure.

Method: The study has been performed by combining a method with structured interviews and questionnaires to all Swedish arenas built between years 2000-2014. A literature study was carried out as foundation of the surveys design, which because of incomplete data had to be supplemented with structured interviews. In order to visualize the Swedish arena-boom and its influencing factors the empirical data was partly analyzed descriptive and partly statistically.

Conclusion: The main reasons behind this study’s arena investments are rationalization and improvement, increased focus on sports, as well as increased social enrichment and enhanced image. During the last decade a trade of ownership structure has taken place and as a result arenas are a majority of the arenas are now privately owned. Statistical analyzes has been made illustrate that the ownership structure doesn’t affected the underlying causes behind the arena investments.

This is a qualitative study that explores the involvement of the relatives in the special housing based on the perspective of fourteen interviewees working in elderly care, at special housing.

The purpose of this study is to analyze the nursing staff's view of the involvement of the relatives and the presence of care provision at special housing. The study is to find out what can constitute support or obstacles to the relatives.

Three questions have been presented as the structural basis upon which the objective finds its conclusion. These questions try to explore and identify the forms of support that can help relatives in their involvement in elderly care. There are opportunities and obstacles for the involvement of the relatives in the care of the elderly as well as expectations of nursing staff on the role of the relatives in the care of the elderly which the following study focuses on.

The research reflected in this study involves the care staff and their experiences and views on the relatives who have their older live permanently in residential care.

In conclusion, the study identifies similar reasoning among respondents regarding to key aspects of the relatives at the special housing. They emphasizes that relatives are needed there and they are important resource for both the residents and the staff.

The results indicate that there may be both opportunities and obstacles for the involvement of the relatives. On one side, factors that prevent relatives from their involvement are lack of communication, poor attitude, different views of relatives and staff about care and conflicts between them. On the other side, there are opportunities that can facilitate relatives' involvement, namely the fact that they need support from the staff. The interviewees pointed out that the relationship between relatives and the elderly is the basis of the relatives' involvement.

This is a qualitative study that explores how bilingual children develop their language through play. The study is based on observations and interviews conducted in a preschool located in a multicultural area of Stockholm.

The purpose of using two qualitative methods; observations and interviews, was to get a bigger picture of the study area. The results of the study, illustrated through observations and interviews that I have conducted, shows that the game has great role for bilingual children's language development. Furthermore, the results show that the informants are aware of the importance of play for bilingual children's language development especially when they are talking about investing in the promotion of children's language by using different methods and materials in their pre activity for various games and activities.

One of my conclusions is that bilingual children learn the language during both free play and structured play especially when educators are present and aware of the activities that they perform. It is also important to see children as individuals to cover all the needs of children at different activities. Finally, I can also mention that bilingual children develop their language in the play by using different materials and communicate in different ways to contact other children. In play, children work a lot with their fantasies, knowledge and experience they already have from before and that they created during the game.

    Abstract [en]

The principal problem that is dealt with in the thesis is to characterize the range of the exponential Radon transform for both constant attenuation and angle dependent attenuation (in the latter case we assume that the attenuation is a trigonometric polynomial). Such results are also of interest in applications such as ECT (Emission Computed Tomography). Abstract [en]

This survey starts with a brief description of the scientific relevance of electron tomography in life sciences followed by a survey of image formation models. In the latter, the scattering of electrons against a specimen is modeled by the Schrödinger equation, and the image formation model is completed by adding a description of the transmission electron microscope optics and detector. Electron tomography can then be phrased as an inverse scattering problem and attention is now turned to describing mathematical approaches for solving that reconstruction problem. This part starts out by explaining challenges associated with the aforementioned inverse problem, such as the extremely low signalto- noise ratio in the data and the severe ill-posedness due to incomplete data, which naturally brings up the issue of choosing a regularization method for reconstruction. Here, the review surveys both methods that have been developed, as well as pointing to new promising approaches. Some of the regularization methods are also tested on simulated and experimental data. As a final note, this is not a traditional mathematical review in the sense that focus here is on the application to electron tomography rather than on describing mathematical techniques that underly proofs of key theorems.

    Abstract [en]

Already in 1968 one recognized that the transmission electron micro- scope could be used in a tomographic setting as a tool for structure determination of macromolecules. However, its usage in mainstream structural biology has been limited and one reason is the devastating combination of noisy data and incomplete data problems that leads to severe ill-posedness of the inverse problem. Despite these issues, the importance of electron tomography is beginning to increase, espe- cially in drug discovery. This review begins with a brief introduction to the model for image formation, i.e. the forward operator. Next, we state the difficulties and review some of the various attempts at overcoming those in solving the inverse problem.

    Abstract [en]

We introduce a reconstruction framework that can account for shape related prior information in imaging-related inverse problems. It is a variational scheme that uses a shape functional, whose definition is based on deformable template machinery from computational anatomy. We prove existence and, as a proof of concept, we apply the proposed shape-based reconstruction to 2D tomography with very sparse and/or highly noisy measurements.

    Abstract [en]

This review of the development and current status of electron tomography deals mainly with the mathematical and algorithmic aspects. After a very brief description of the role of electron tomography in structural biology, we turn our attention to the derivation of the forward operator. Starting from the Schrodinger equation, the electron - specimen interaction is modelled as a diffraction tomography problem and the picture is completed by adding a description of the optical system of the transmission electron microscope. The first- order Born approximation enables one to explicitly express the intensity for any finite wavenumber in terms of the propagation operator acting on the specimen convolved with a point spread function, derived from the optics in the transmission electron microscope. Next, we focus on the difficulties that cause the reconstruction problem to be quite challenging. Special emphasis is put on explaining the extremely low signal- to- noise ratio in the data combined with the incomplete data problems, which lead to severe ill- posedness. The next step is to derive the standard phase contrast model used in the electron tomography community. The above- mentioned expression for the intensity generalizes the standard phase contrast model which can be obtained by replacing the propagation operator by its high- energy limit, the x- ray transform, as the wavenumber tends to infinity. The importance of more carefully including the wave nature of the electron - specimen interaction is supported by performing an asymptotic analysis of the intensity as the wavenumber tends to infinity. Next we provide an overview of the various reconstruction methods that have been employed in electron tomography and we conclude by mentioning a number of open problems. Besides providing an introduction to electron tomography written in the 'language of inverse problems', the authors hope to raise interest among experts in integral geometry and regularization theory for the mathematical and algorithmic difficulties that are encountered in electron tomography.

We present a new local tomographic algorithm applicable to electron microscopy tomography. Our algorithm applies to the standard data acquisition method, single-axis tilting, as well as for more arbitrary acquisition methods. Using microlocal analysis we put the reconstructions in a mathematical context, explaining which singularities are stably visible from the limited data given by the data collection protocol in the electron microscope.

    Abstract [en]

We present a new local tomographic algorithm applicable to electron microscope tomography. Our algorithm applies to the standard data acquisition method, single-axis tilting, as well as to more arbitrary acquisition methods including double axis and conical tilt. Using microlocal analysis we put the reconstructions in a mathematical context, explaining which singularities are stably visible from the limited data given by the data collection protocol in the electron microscope. Finally, we provide reconstructions of real specimens from electron tomography data.

    Abstract [en]

Filtered back-projection and weighted back-projection have long been the methods of choice within the electron microscopy com- munity for reconstructing the structure of macromolecular assem- blies from electron tomography data. Here, we describe electron lambda-tomography, a reconstruction method that enjoys the ben- efits of the above mentioned methods, namely speed and ease of implementation, but also addresses some of their shortcomings. In particular, compared to these standard methods, electron lambda- tomography is less sensitive to artifacts that come from structures outside the region that is being reconstructed, and it can sharpen boundaries.
