Adipogenic Differentiation Potential of Stem Cells Under The Influence of the Epigenetic Modifiers 5-Aza-2´-deoxycytidine and Suberoylanilide hydroxamic acid

Patients that have gone through mastectomy and burn victims have high psychological burden while current solutions are associated with complications. The mesenchymal stem cells (MSC) have the ability to differentiate into many cell types including adipocytes. Adipocytes can be found throughout the human body but also in loose connective tissue mixed with other cell types. In this study MG63 cell line was used as it has the characteristics of MSC. The epigenetic modifiers DNA methylation inhibitor 5-Aza-2´-deoxycytidine (5-Aza-dC) and the histone deacetylase inhibitor Suberoylanilide hydroxamic acid (SAHA) were studied to investigate their effect on the adipogenic differentiation protocol. SAHA increased while 5-Aza-dC decreased fat accumulation in comparison to the control group and vehicle as shown by the oil red staining. The gene expression showed the same results for the following genes: peroxisome proliferator-activated receptor gamma, adiponectin receptor I and II and for glucose transporter type 4. Compared to the control, the adipogenic inducer; monocyte chemotactic protein-1 secretion reached up to 165% for SAHA and to less than 22% for 5-Aza-dC in the supernatant culture. The study shows SAHAs potential in the fat regeneration and also 5-Aza-dC potential role as an anti-obesity agent.
